Antioxidant and Protective Effects of a Standardized Chamaecrista nomame (Siebold) H. Ohashi Extract against Ultraviolet B in Human Dermal Fibroblasts; [표준화된 차풀 추출물의 항산화 및 UVB로 손상된 피부 섬유아세포 보호 효과]

This study sought to investigate the antioxidant activity of a standardized extract of Chamaecrista nomame (Siebold) H. Ohashi (CE) and its protective effects against ultraviolet B (UVB)-induced skin cell damage. Antioxidant capacity was assessed using 2, 2′-azino-bis (3-ethylbenzthiazoline-6-sulfonic acid) (ABTS), ferric reducing antioxidant potential (FRAP), reducing power, and oxygen radical absorbance capacity (ORAC) assays. The cell protective effect was performed in human dermal fibroblasts (HDF) using by cytotoxicity test, and recovery of cell viability was determinded using UVB irradiation. The intercellular reactive oxygen species (ROS) after UVB irradiation were analyzed using 2′, 7′-dichlorofluorescin diacetate. In the ABTS assay, the CE showed a concentration-dependent increase in radical scavenging activity at 100, 200, and 300 μg/mL. FRAP and reducing power also increased in treatment concentration, indicating that CE has the ability to reduce ferric iron (Fe3+) to ferrous iron (Fe2+). The ORAC value was calculated using the trolox calibration curve, which was 209.38±6.00 µM trolox equivalent. In the cell-based assays, the extract exhibited no cytotoxicity at 100∼300 µg/mL and restored the HDF cell viability reduced by UVB. Also UVB-elevated intercellular ROS levels decreased in a concentration-dependant manner. These results indicate that CE shows antioxidant and skin-protective effects. These findings supports its potential as a functional food and inner beauty food ingredient. © 2025 The Korean Society of Food Science and Nutrition.
