The expression of many genes regulates organogenesis during somatic embryogenesis. Since the Myb gene is known to be involved in diverse cellular functions, we isolated the Myb gene from Pimpinella brachycarpa and analyzed its expression during somatic embryogenesis of P. brachycarpa. Somatic embryos were easily obtained from suspension culture of leaflets on MS medium containing 0.2 μM 2,4-D, 0.5 μM BAP and 10.0 μM TDP. The isolated Myb gene from P. brachycarpa, Pbmyb, was 2,439 bp long and contained one intron of 1,200 bp. The Pbmyb coded 269 amino acids and contained a highly conserved DNA binding domain in its N-terminal region and two incompletely duplicated sequences. Southern hybridization showed four copies of the Pbmyb, which is very low compared to other plants. The Pbmyb was expressed in calli, shoots and in vitro plants. In checking tissue specificity of the expression, the expression of the Pbmyb was observed in flowers, leaves and roots. Therefore, we propose that expression of the Pbmyb gene might be related with organogenesis rather than embryogenesis during somatic cell differentiation.
