Determination of polychlorinated biphenyl 126 in liver and adipose tissues by GC-μECD with liquid extraction and SPE clean-up

Simple, rapid, accurate, and reproducible gas chromatographic methods were developed for the quantitative determination of PCB 126 in liver and adipose tissues. PCB 126 was extracted from the tissues by homogenizing the former with hexane and by alkaline hydrolysis of the latter with 2 N NaOH solution, respectively. Sample clean-up was conducted employing the Varian Bond Elut (R) PCB cartridge, using hexane as an eluant. The developed methods were validated for calibration curve linearity, accuracy, precision, and lower limit of quantitation. The applicability of these methods can be expanded to the analyses of other PCB congeners in similar biological tissues.
