돌연변이 Saccharomyces cerevisiae YJS17Mal 균주를 이용한 전분의 동시당화발효

The acceleration of global warming has prompted a significant focus on starch-based bioethanol as a potential alternative to gasoline. The production of starch-based bioethanol involves SSF (simultaneous saccharification and fermentation) of starch and it is preferable to use yeast strains that can ferment maltose, a breakdown product of starch. In this study, 16 strains of Saccharomyces cerevisiae were isolated from a forest in Kangwon, Korea. Of the three S. cerevisiae strains that gained maltose metabolism ability after maltose enrichment culture, S. cerevisiae YJS17Mal was studied further. When comparing the maltase enzyme activity from the S. cerevisiae YJS17Mal and its parent strain, it was confirmed that the extract from the S. cerevisiae YJS17Mal could degrade maltose. The gene and amino acid sequences were compared, but no differences were found. SSF of starch was performed using both strains using 1500 U/L of α-amylase. S. cerevisiae YJS17Mal produced 9.08 g/L of ethanol. S. cerevisiae YJS17Mal was confirmed as suitable candidate for SSF of starch, as analysis revealed the absence of residual starch at the end of the process.
