초록
Cancer affects millions of people worldwide and is the second leading cause of death after heart disease. Since the latter half of the last century, the collaboration among biology, chemistry, engineering, and medicine has highlighted the application of nanotechnology in cancer treatment. Particularly, lipid-based drug delivery systems (LBDDS) with biodegradability and biocompatibility have garnered significant attention in cancer therapy. These systems improve the efficiency and safety of drug delivery by considering the tumor microenvironment (TME), offering advantages such as targeted delivery, controlled release, and extended circulation time. LBDDS have been developed in various forms, including liposomes, microemulsions, solid lipid nanoparticles, and nanostructured lipid carriers. These systems can carry both hydrophobic and hydrophilic drugs, extend the half-life of drugs to prolong their action, and reduce toxicity. The success of FDA-approved LBDDS like Doxil and Paclitaxel demonstrates the potential of nanoparticle-based therapies. This review discusses the various types of lipid-based drug delivery systems and their mechanisms of cancer targeting, encompassing a wide range of application examples from recent research trends. Through this, it brings about innovation in cancer treatment methods and is expected to play a crucial role in future cancer therapies.
