We have investigated the surface plasmon resonance (SPR) angle shift due to the interaction of a monoclonal anti-C-reactive protein (CRP) with a CRP on surface-modified protein arrays by using an angular interrogation-based SPR sensor. The surface structure of the protein arrays consisted of glass/metal/proteins/ air (ex situ). The SPR sensor was self-constructed, and its detection limit was calculated to be 4.6 x 10(-5). The SPR angle monotonically increased with increasing CRP concentration from 1 to 100 mug/ml and pronouncedly increased after interaction of monoclonal anti-CRP and CRP on the protein arrays, which could be explained by the changes in the refractive index of the gold surface of the protein arrays. These results suggest that ex situ SPR. sensors can be used as immunosensors for the analysis of protein arrays.
