In this study, as part of the standardization of raw materials for the development of a Ziziphus jujuba Mill.-Hypericum perforatum L. mixture as a functional ingredient, a high-performance liquid chromatography analytical method was validated for betulinic acid and pseudohypericin, the indicator substances in this mixture. The validation was performed by evaluating specificity, linearity, accuracy, precision, limit of detection (LOD), and limit of quantification (LOQ). The coefficients of determination (R2) for the calibration curves of betulinic acid and pseudohypericin were 1 and 0.9993, respectively, indicating high linearity. The LOD/LOQ of betulinic acid was 0.14/0.42 μg/mL, and that of pseudohypericin was 0.07/0.22 μg/mL. The accuracy test at three concentration levels confirmed the accuracy of both analytes, with 80-120% accuracy. In addition, the precision test results of both betulinic acid and pseudohypericin showed less than 5%. Ziziphus jujuba Mill.-Hypericum perforatum L. mixture showed excellent antioxidant activity, as measured by 2,2′-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) radical scavenging assay, ferric reducing antioxidant power, and oxygen radical absorbance capacity assays. The results of this study provide basic data for the standardization of the Ziziphus jujuba Mill.-Hypericum perforatum L. mixture and suggest its potential use as a functional food and as a natural antioxidant material. © 2026 The Korean Society of Food Science and Nutrition.
