Temperature- and oxidation-dependent doxorubicin release from poly(hydroxyethyl acrylate-co-phenyl vinyl sulfide) cryogel

We synthesize temperature- and oxidation-responsive poly(hydroxyethyl acrylate-co-phenyl vinyl sulfide) (P(HEA-co-PVS)) cryogel for the delivery of doxorubicin (DOX). We observed that the cloud point temperature (CPT) of P(HEA-co-PVS) copolymer decreased with increasing PVS. The NIR treatment increased the temperature of the polymer above its CPT, resulting in the dissolution of the polymer. With increasing PVS content, the minimum interfacial activity of the copolymers was increased, indicating increase in their amphiphilicity. The H2O2 treatment oxidized the sulfide group of PVS reducing the hydrophobicity of the copolymer, decreasing its amphiphilicity, and releasing the drug. The SEM micrographs showed a continuous polymer matrix cryogel produced from P(HEA-co-PVS). The cryogel showed a more swelling ratio when the PVS content is less due to less intermolecular hydrophobic interaction. The release of DOX was responsive to the temperature, ROS, and NIR irradiation, suggesting that the cryogels could be a suitable anticancer drug delivery carrier to tumor environments.
