We have investigated whether wavelength interrogation-based surface plasmon resonance (SPR) sensors are able to analyze immunoreactions on protein arrays. The surface formation of protein arrays (Au/mixed thiol layer/proteins/air) was analyzed by using the shift of resonance wavelength and atomic force microscopy. The specific interaction of Rac1 with various concentrations of Rac1 antibody was successfully analyzed on the protein arrays and was explained by using the changes in the refractive index of the protein arrays. These results suggest that the wavelength interrogation-based SPR, sensors can be used as immunosensors for high-throughput analysis of antigen-antibody interactions on protein arrays.
