Antibacterial activity of ethanol extracts from house fly-maggot against various strains of Methicillin-resistant Staphylococcus aureus and Vancomycin-resistant enterococci strain

초록

In a previous study, we reported the existence of a 5 kDa peptide in the water-soluble protein hydrolysates obtained from fly maggots that inhibits Methicillin-resistant Staphylococcus aureus (MRSA). In the present successive study, the antibacterial activity of the hydrolysates and the ethanol (EtOH)-extracted fraction against MRSA and Vancomycin-resistant enterococci (VRE) 5117 was determined. Hydrolysates were prepared by a 4 or 8 h enzyme treatment followed by thermolysin treatment. EtOH extracts A (uniform extract at room temperature), B (extracted for 24 h at 4oC) and C (extracted for 24 h at -20oC) were also obtained. The growth curves for MRSA strain 3595 and VRE strain 5117 showed more potent and concentration-dependent inhibition by the 8 h hydrolysates as compared with the 4 h hydrolysates. MRSA and VRE inhibition was highest for EtOH extract C. The minimum inhibitory concentration of extract C against MRSA strains 3589, 3595, 3596, 3597, 3598 and 3601 was 60, 60, 50, 40, 40 and 50 μg/ml, respectively. While the butanol fraction from EtOH extract C displayed potent antibacterial effect against MRSA and VRE strains, the antibacterial effects of the hexane, ethyl acetate, and water layers could not be confirmed.
