Cell membrane-based nanovaccines have demonstrated attractivefeaturesdue to their inherently multiantigenic nature and ability to be formulatedwith adjuvants. Here, we report on cellular nanodiscs fabricated fromcancer cell membranes and incorporated with a lipid-based adjuvantfor antitumor vaccination. The cellular nanodiscs, with their smallsize and discoidal shape, are readily taken up by antigen-presentingcells and drain efficiently to the lymph nodes. Due to its highlyimmunostimulatory properties, the nanodisc vaccine effectively stimulatesthe immune system and promotes tumor-specific immunity. Using a murinecolorectal cancer model, strong control of tumor growth is achievedin both prophylactic and therapeutic settings, particularly in combinationwith checkpoint blockades. Considerable therapeutic efficacy is alsoobserved in treating a weakly immunogenic metastatic melanoma model.This work presents a new paradigm for the design of multiantigenicnanovaccines that can effectively activate antitumor immune responsesand may be applicable to a wide range of cancers.
