Efficacy of antibacterial and antibiofilm of silver nanoparticles decorated multi-walled carbon nanotubes loaded PF127 hydrogel

Silver nanoparticles (AgNPs) have been suggested as antibacterial agents against multi-drug-resistant bacterial pathogens, although their toxicity concerns necessitate refinement before their application in wound care. Hence, this study investigates AgNPs-decorated multi-walled carbon nanotubes (MWCNT-Ag) loaded pluronic F127 (PF127) hydrogel composites for potential use in wound management. The prepared AgNPs, MWCNT-Ag, and PF127 formulations were characterized by dynamic laser scattering, electrophoretic light scattering, Fourier transform infrared spectroscopy, X-ray diffraction, transmission electron microscopy, and scanning electron microscopy. At room temperature, AgNPs/PF127 and MWCNT-Ag/PF127 remained in a solution state and exhibited a gel transition at 37 degrees C. The viscosity of PF127 hydrogel was increased with the addition of AgNPs and MWCNT-Ag. MWCNT-Ag demonstrates comparable or enhanced antibacterial efficacy compared to AgNPs against both Gram-positive and Gram-negative bacteria. The MIC90 of AgNPs was 272 to 447 mu g/mL, while the MIC90 of MWCNT-Ag ranged from 120 to 467 mu g/mL. The antibacterial and biofilm inhibition properties of PF127-based hydrogel composites are shown to be improved through synergistic interactions. The PF127 formulation exhibited significant antioxidant activity against DPPH and ABTS, exhibiting approximately 70 % ABTS radical inhibition at 31.2 mu g/mL. Furthermore, in vitro cytotoxicity assessment reveals increased cell viability with MWCNT-Ag composites compared to AgNPs. In the wound healing study, the average scratch closure for control, AgNPs/PF127, and MWCNT-Ag/PF127 at 36 h was 72 %, 89 %, and 99 %, respectively. In conclusion, these findings suggest that PF127 hydrogel composites present a promising avenue for drug delivery in wound healing, potentially mitigating toxicity concerns.
