We report the preparation and cytotoxicity of TiO2 nanoparticles (TiO2 NPs) using black cumin sprout extract. The average size of BC-TiO2 NP was 97.48 +/- 1.54 d. nm, with a PDI of 0.31, and zeta potential of-29.1 +/- 0.89 mV. The size of NPs varied between the methods (TEM, XRD, and DLS) due to the hydrodynamic properties of BC-TiO2 NPs. The cytotoxicity studies evidenced that BC-TiO2 NPs exhibited a half maximum inhibitory con-centration (IC50) of 76.25 +/- 1.57 mu g/mL for the human lung cancer cell line (A549) and 175.64 +/- 3.48 mu g/mL for the mouse fibroblast cell line (NIH3T3). In addition, BC-TiO2 NPs triggered the antiproliferative activity in the A549 cell line through nucleus damage, mitochondrial membrane potential loss, and apoptosis-associated membrane changes while showing no toxic effects on egg CAM and RBC. These results revealed that BC-TiO2 NPs exhibited strong cytotoxicity in the A549 cell line while being nontoxic to the NIH3T3 cell line, egg CAM, and RBC.
