Complete genome sequence of a potato virus Y isolated from Browallia speciosa in Korea

In 2024, a Browallia speciosa plant exhibiting necrotic spots and vein clearing symptoms on the leaves was collected in Chuncheon, Korea. High-throughput sequencing (HTS) was performed on total RNA extracted from the B. speciosa leaves, generating 30,733,016 reads and assembling 10,230 contigs through de novo assembly. Among these, 264 contigs mapped to potato virus Y (PVY) of the genus Potyvirus. The genome displayed a typical PVY organization, consisting of a single open reading frame (ORF) encoding a polyprotein. By comparison with previously reported cleavage sites, the positions and lengths of ten mature peptides and the PIPO coding sequence (CDS) were determined. RT-PCR was performed using the total RNA, and PVY was detected. Phylogenetic analysis based on the complete genome and polyprotein regions indicated that the B. speciosa isolate of PVY belongs to the NTNa strain. This isolate shared 99.73% nucleotide identity with a previously reported Korean PVY-NTNa isolate (MH603862.1). This study represents the first report of a complete genome sequence of a PVY isolate infecting B. speciosa plant in Korea. Copyright ⓒ 2026, The Microbiological Society of Korea.
