Genetic diversity and population structure of alder, Alnus japonica using AFLP markers

The genetic diversity and population structure among 17 natural populations of alder, Alnus japonica Thunb, were estimated using amplified fragment length polymorphism (AFLP). A total of 360 DNA fragments were scored. Although some Korean populations were isolated and patchily distributed, they exhibited a high level of genetic diversity. On average, 49.4% of AFLP markers generated using eight primer pairs were polymorphic. The combination of a wind-pollinated, outcrossing breeding system along with large population sizes, and the ability to regenerate by stump sprouting may explain the high level of genetic diversity within this species. Most of the genetic variance of alder (94.8%) resided within populations. The southern populations of A. japonica with small sites were isolated from each other, being surrounded by the introduced alder (A. firma) in the same regions.
