Ideonella sakaiensis 유래 재조합 PETase의 생산 조건 최적화 및 PETase 활성 분석

초록
The environmental problem caused by increasing plastic waste is becoming serious. As a method for recycling this waste, using enzymes to convert polyethylene terephthalate (PET) into its monomer has emerged without side effects. Among various enzymes, PETase has the characteristic of being able to degrade PET more effectively even at room temperature, unlike other enzymes; however, its application in industrial application is still insufficient. To address this, an expression system for producing recombinant PETase was established in this study. Through the optimization of expression conditions, 0.4 mg/mL of recombinant PETase was produced. Subsequently, the degradation of PET by the recombinant PETase was confirmed through FE-SEM and FT-IR analysis. Furthermore, the concentration of terephthalic acid (TPA), the degradation product of PET by the recombinant PETase, was measured using HPLC. This analysis confirmed the highest TPA release (16.23 mg/L) under the reaction condition of 25°C. Through research on optimizing the production and reaction conditions of recombinant PETase, it is expected to contribute to the preservation of the global environment.
