Characterization of a novel ion pair with temperature and oxidation response capabilities and its application in liposomes

An ion pair containing poly (ethylenemine) (PEI) and 4- (Methylthio) phenylacetic acid (MTPA) was prepared to achieve temperature and oxidation sensitivity and used as a helper molecule in the preparation of dioleoylphysidylethanolamine (DOPE) liposomes. The behavior of upper critical solution temperature (UCST) can be observed in the PEI/MTPA ion pair and decreased as the concentration of hydrogen peroxide (H2O2) increased. And nanosized spherical morphology of PEI/MTPA captured by transmission electron microscopy (TEM) images. The results of FT-IR and 1H NMR exhibited the formation of salt bridges and proton peak shifts due to the interaction between amino and carboxyl groups. MTPA in ion pair was confirmed to be oxidized by H2O2 through XPS results. In addition, due to the amphiphilic property of the PEI/MTPA ion pair, the DOPE liposome bilayer can be stabilized by the ion pair. Compared with the control, liposomes containing PEI/MTPA ion pairs released their payload under heating and oxidation conditions. The cell viability result indicated that the prepared liposomes have no cytotoxicity to NIH3T3 cells and can be used as biomaterials.
