A Fiber-Optic Localized Surface Plasmon Resonance (FO LSPR) sensor, which is fabricated using gold nanoparticles immobilized on the surface of the optical fiber, can be used as a label-free immunoassay biosensor. In the measurement of an immunoassay using the FO LSPR sensor, it is important to optimize the immobilization of antibodies on the surface of gold nanoparticles, in order to obtain larger signal from the specific bindings of antigens against antibodies. In this paper, we propose the methods of optimization for immobilizing antibodies to get the performance improvement of FO LSPR biosensor. To verify the proposed methods, various experiments were implemented with the changes of experimental conditions such as the dipping time and the number of the dipping into the antibody solution in Prostate Specific Antigen (PSA) immunoassay. Also, we proposes the in-situ method to analyze the antibody immobilization, and the proposed analysis method has been evidenced by using measured sensorgrams of the PSA immunoassay.
