Protein arrays are a promising technology for the high-throughput analysis of molecular interactions, protein profiling, and the identification and evaluation of protein biomarkers with only very small amounts of samples. With increasing interest in the functions and roles of enzymes in biological systems, enzymatic profiling based on protein chip technology has emerged as an alternative for the characterization of enzymes in various biological processes. In this review, we describe on-chip activity assay strategies, quantification methods for enzymatic profiling, and applications of activity-based protein chips for enzymatic profiling and serodiagnosis.
